Evaluating molecular diagnostic techniques for seed detection of Pseudomonas savastanoi pv. phaseolicola, causal agent of halo blight disease in mungbean (Vigna radiata)

چکیده

Abstract Halo blight of mungbean ( Vigna radiata var. ) is caused by the bacterium Pseudomonas savastanoi pv. phaseolicola . This pathogen transmitted via infected seed, facilitating spread disease into new cultivated areas. Prospective seed crops are currently subjected to visual inspection as a means determining status, however, this poor method that relies on visible symptoms and does not account for latent infections. A range molecular diagnostics targeting P. have been developed, but these deployed seeds. Quantitative PCR (qPCR) SYBR assay, hydrolysis probe, conventional PCR, using same primers were optimised against plate-truthed dilution series The detection limit assay was approximately 9,000 CFU µl -1 , while both qPCR assays could detect 9 These tests then used screen DNA extracted from 200 g allotments 38 lots comprising six cultivars representing primary Australian production area, two known infection status. Of these, detected in PCR. probe methods 20 24 respectively. shows most effective at diagnosing presence providing valuable diagnostic aid integrated management certification, substantially mitigating losses halo disease.

Occurrence of Pseudomonas savastanoi the Causal Agent of Winter Jasmine Gall in Iran

During the years 2007 and 2008, samples of winter jasmine (Jasminum nudiflorum) with symptoms of galls on shoots were collected from Shiraz, Fars province, Iran. Gram and oxidase negative, aerobic bacterium that produced fluorescent pigment on Kingۥs B medium was isolated from galls of infected trees. All isolates produced a hypersensitive reaction in tobacco. They were negative in arginine dih...

Genetic Confirmation of Mungbean (Vigna radiata) and Mashbean (Vigna mungo) Interspecific Recombinants using Molecular Markers

Molecular confirmation of interspecific recombinants is essential to overcome the issues like self-pollination, environmental influence, and inadequacy of morphological characteristics during interspecific hybridization. The present study was conducted for genetic confirmation of mungbean (female) and mashbean (male) interspecific crosses using molecular markers. Initially, polymorphic random a...

Endopathogenic lifestyle of Pseudomonas savastanoi pv. savastanoi in olive knots

The endophytic phase of Pseudomonas savastanoi pv. savastanoi in olive stems and the structural and ultrastructural histogenesis of olive knots have been studied. Construction of a stable plasmid vector expressing the green fluorescent protein, in combination with the use of in vitro olive plants, allowed real-time monitoring of P. savastanoi pv. savastanoi infection. The infection process was ...

Bacterial blight (Xanthomonas arboricola pv.juglandis) is one of the main diseases of walnut that reduce the yield in the central, western and northern regions of Iran. This disease was first reported form, Qazvin and Mazandaran, then widely reported from northern, central and western provinces. To identify the cause of the disease in different provinces, infected leaves collected from Alborze,...

Seed Yield of Mungbean (Vigna radiata (L.) Wilczek) in relation to Growth and Developmental Aspects

Growth parameters such as leaf area (LA), total dry mass (TDM) production, crop growth rate (CGR), relative growth rate (RGR), and net assimilation rate (NAR) were compared in six varieties of mungbean under subtropical condition (24°8' N 90°0' E) to identify limiting growth characters for the efficient application of physiology breeding for higher yields. Results revealed that a relatively sma...
